{"id":67775,"date":"2025-02-11T16:50:19","date_gmt":"2025-02-11T05:50:19","guid":{"rendered":"https:\/\/hudson.org.au?post_type=hdsn_news_post&#038;p=67775"},"modified":"2025-02-12T17:28:02","modified_gmt":"2025-02-12T06:28:02","slug":"genetic-analysis-caps-search-for-a-cure","status":"publish","type":"hdsn_news_post","link":"https:\/\/hudson.org.au\/news\/genetic-analysis-caps-search-for-a-cure\/","title":{"rendered":"Genetic analysis CAPS search for a cure"},"content":{"rendered":"\n<p class=\"has-medium-font-size\">A rare and debilitating group of autoinflammatory diseases is closer to a cure thanks to a world first genetic analysis project at Hudson Institute.<\/p>\n\n\n\n<p>Cryopyrin-associated periodic syndromes (CAPS) are inherited autoinflammatory conditions characterised by recurrent bouts of systemic inflammation related to inappropriate activation of the innate immune system.<\/p>\n\n\n\n<p>These diseases are known to be linked to genetic changes in an immune senser named NLRP3 and now, for the first time, researchers have analysed hundreds of mutations to pinpoint the causes.<\/p>\n\n\n\n<p>Their work has been published in the journal <em><a href=\"https:\/\/www.nature.com\/articles\/s41590-025-02088-9\" target=\"_blank\" rel=\"noreferrer noopener\">Nature Immunology<\/a><\/em>.<\/p>\n\n\n\n<p>First author, <a href=\"https:\/\/hudson.org.au\/researcher-profile\/shouya-feng\/\">Dr Shouya Feng<\/a> said this research is the world-first functional screening of all known NLRP3 variants: \u201cThis extensive analysis provides the first systematic approach to understanding how specific mutations lead to inflammasome activation.\u201d<\/p>\n\n\n\n<h2 class=\"wp-block-heading\" id=\"h-first-ever-caps-genetic-analysis\">First ever CAPS genetic analysis<\/h2>\n\n\n\n<p>Her supervisor, <a href=\"https:\/\/hudson.org.au\/researcher-profile\/seth-masters\/\">Professor Seth Masters<\/a>, said most of these gene variants have never been studied before.<\/p>\n\n\n\n<p>\u201cOur analysis shows that some people got diagnosed with this condition incorrectly, and there are quite a number of people out there whose disease didn\u2019t look exactly like CAPS, but it turns out our test shows that this is what they have, and for the first time they can get access to a drug that treats the condition, insurance coverage, family planning and certainty about why they are ill,\u201d Prof Masters said.<\/p>\n\n\n<div class=\"wp-block-image\">\n<figure class=\"alignright size-medium\"><a href=\"https:\/\/hudson.org.au\/wp-content\/uploads\/2025\/02\/nlrp3-colour-image-shouya-and-seth-nature-paper.jpg\"><img loading=\"lazy\" decoding=\"async\" width=\"300\" height=\"300\" src=\"https:\/\/hudson.org.au\/wp-content\/uploads\/2025\/02\/nlrp3-colour-image-shouya-and-seth-nature-paper-300x300.jpg\" alt=\"Distribution of highly active (red) and inactive (blue) variants on NLRP3 structure based on our research.\" class=\"wp-image-67777\" srcset=\"https:\/\/hudson.org.au\/wp-content\/uploads\/2025\/02\/nlrp3-colour-image-shouya-and-seth-nature-paper-300x300.jpg 300w, https:\/\/hudson.org.au\/wp-content\/uploads\/2025\/02\/nlrp3-colour-image-shouya-and-seth-nature-paper-150x150.jpg 150w, https:\/\/hudson.org.au\/wp-content\/uploads\/2025\/02\/nlrp3-colour-image-shouya-and-seth-nature-paper.jpg 600w, https:\/\/hudson.org.au\/wp-content\/uploads\/2025\/02\/nlrp3-colour-image-shouya-and-seth-nature-paper-320x320.jpg 320w, https:\/\/hudson.org.au\/wp-content\/uploads\/2025\/02\/nlrp3-colour-image-shouya-and-seth-nature-paper-400x400.jpg 400w\" sizes=\"auto, (max-width: 300px) 100vw, 300px\" \/><\/a><figcaption class=\"wp-element-caption\"><em>Distribution of highly active (red) and inactive (blue) variants on NLRP3 structure.<\/em><\/figcaption><\/figure>\n<\/div>\n\n\n<p>Dr Feng and the team studied more than 500 genetic variants of NLRP3 to see how they affect inflammasome activation and response to inhibitors.<\/p>\n\n\n\n<p>\u201cOur findings reveal different ways these mutations trigger inflammation, identify variants resistant to existing drugs, and provide new insights for diagnosing and treating patients with CAPS. This work could help match patients to the right treatments and improve future drug development,\u201d she said.<\/p>\n\n\n\n<p>Dr Feng believes that improving the classification of pathogenic and benign NLRP3 variants will ensure that patients can receive the right care.<\/p>\n\n\n\n<p>\u201cAdditionally, as new NLRP3-targeting drugs like MCC950-derived compounds are being developed, our findings provide valuable insights to improve drug design and expand treatment options for those with resistant mutations,\u201d she said.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\" id=\"h-disease-facts-caps\">Disease facts &#8211; CAPS<\/h2>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Cryopyrin-Associated Periodic Syndromes (CAPS) are rare autoinflammatory diseases caused by NLRP3 mutations, affecting approximately 3 to 5 in 1 million people worldwide.<\/li>\n\n\n\n<li>CAPS comprises the three conditions previously described as familial cold autoinflammatory syndrome (FCAS), Muckle-Wells syndrome (MWS), and neonatal-onset multisystem inflammatory disorder (NOMID), also known as chronic infantile neurologic, cutaneous, and articular (CINCA) syndrome.<\/li>\n\n\n\n<li>General symptoms are fatigue and fever.
